Has anyone else been on Avmapki and Fakzynja?

This is a newly approved combination treatment (approved in 2025), so it's still quite new. You may not find many members who have personal experience with it yet, but as more people start this therapy, experiences will be shared.

What is this medication for?

Avmapki and Fakzynja (avutometinib and defactinib) work together to block signals that help cancer cells grow and survive. This combination is specifically approved for adults with KRAS-mutated, low-grade serous ovarian cancer that has returned after earlier treatment. It's actually the first targeted treatment specifically designed for this rare and hard-to-treat form of ovarian cancer.

How does it work?

These two oral medications are dual kinase inhibitors that target certain pathways cancer cells use to survive and multiply.

What should you know?

Your doctor can test your tumor for KRAS mutations to confirm if this treatment is right for you. In clinical trials, some people responded well to this combination, though more research is ongoing to understand long-term benefits. Like other targeted therapies, it may not be suitable for everyone.

Consider discussing any questions or concerns about side effects and what to expect with your oncology team.

I’m starting my 3rd month on this combo. The side effects are manageable sometimes. I live on Imodium for gastrointestinal issues. Fatigue is another issue. I take it for 3 weeks and then a week off. Was able to enjoy our cruise to St Thomas last week. Weather was great. Came home to cooler temperatures. But it’s Florida so it will pass quickly.
